Detection Principle
线粒体呼吸链复合体IV能够催化还原型细胞色素C生成氧化型细胞色素C，还原型细胞色素C在550 nm处具有特征吸收峰，通过吸光值的下降速率即可表征线粒体复合体IV的活性。

①若A1测定大于1.0或∆A大于0.2，建议将粗酶液适当稀释后再进行测定，计算时相应修改；
②提取液中含有约1 mg/mL的蛋白，测定样本蛋白浓度时需减去提取液自身的蛋白含量；
③准确在10 s和70 s处完成吸光值的测定，以确保实验结果的准确性和重复性；若使用96孔板应使用多道移液器且分批进行测定，以确保组间反应时间一致；
注： 为保证结果准确且避免试剂损失，测定前请仔细阅读说明书（以实际收到说明书为准），确认试剂储存和准备是否充分，操作步骤是否清楚，且务必取2-3个预期差异交的样本进行预测定，过程中问题请您及时与工作人员联系。
