Detection Principle
JC-1是一种广泛用于检测线粒体膜电位∆Ψm的理想荧光探针,可以检测细胞、组织或纯化的线粒体膜电位。在线粒体膜电位较高时,JC-1聚集在线粒体的基质中形成聚合物,可以产生红色荧光;在线粒体膜电位较低时,JC-1不能聚集在线粒体的基质中,此时JC-1为单体,可以产生绿色荧光。通过荧光颜色的转变即可检测线粒体膜电位的变化。

Notices

①JC-1(200×)低温条件下可能出现凝固,建议25℃水浴孵育至完全融解后使用;

②JC-1探针装载完成后需置于冰上低温放置,并在30 min内完成检测;

③若JC-1 Dyeing Buffer(5×)中出现沉淀,建议37℃水浴孵育至完全融解后使用;

④CCCP为线粒体电子传递链抑制剂,具有一定毒性,请注意做好防护;

注: 为保证结果准确且避免试剂损失,测定前请仔细阅读说明书(以实际收到说明书为准),确认试剂储存和准备是否充分,操作步骤是否清楚,且务必取2-3个预期差异交的样本进行预测定,过程中问题请您及时与工作人员联系。
