Detection Principle
土壤硝酸还原酶催化硝酸盐还原为亚硝酸盐，NO3-+NADH+H+→NO2-+NAD++H2O，亚硝酸盐能够在酸性条件下，与对-氨基苯磺酸及α-萘胺生成红色偶氮化合物，未反应的NADH会抑制后续的显色反应，使用PMS将其中和后再进行后续反应，红色偶氮化合物在520 nm处具有特征吸收峰，通过吸光值变化即可表征土壤硝酸还原酶的活性。

①试剂一、试剂二和试剂四需冰上放置，使用完成后及时置于-20℃分装保存，避免反复冻融；
②若测定吸光值超出标准线性范围：高于最高值建议将上清液使用蒸馏水稀释后或适当调整样本量后再进行测定，低于最低值可适当延长第一次37℃反应时间后再进行测定，计算时相应修改；
注： 为保证结果准确且避免试剂损失，测定前请仔细阅读说明书（以实际收到说明书为准），确认试剂储存和准备是否充分，操作步骤是否清楚，且务必取2-3个预期差异交的样本进行预测定，过程中问题请您及时与工作人员联系。
