Detection Principle
脲酶水解尿素产生的NH3-N,在强碱性介质中能够与次氯酸钠和苯酚反应,生成水溶性蓝色染料靛酚蓝,产物在630 nm处具有特征吸收峰,通过吸光值的变化即可表征土壤脲酶的活性。

Notices

①若测定吸光值超出标准吸光值线性范围:高于最高值建议适当扩大上清液稀释倍数后再进行测定,低于最低值建议适当增加样本量后再进行测定,计算时相应修改;

②显色完成后应在1 h内完成测定,测定组颜色应为蓝色(与标准组颜色一致),若显色为黄色或绿色,则表明NH3-N浓度过高,调整样本量或上清稀释液稀释倍数后再进行测定,计算时相应修改;

③无土样组为基质自分解对照组,测定组上清液稀释倍数不变则无土样组只需测定1-2次,若改变上清液稀释倍数则需要重新测定无土样组1-2次(稀释倍数一致);
